Can Your HPLC Column Do This? Water Soluble Vitamins on Cogent Bidentate C18 with TYPE-C Silica Page 30 Many generic pharmaceutical companies manufacture vitamins. But the "Water Soluble Vitamin" content analysis is still not a fully routine HPLC method, as these target compounds are very polar and, therefore not well retained on standard type B columns. Their analysis often requires a large "ion pair" in the eluent to facilitate retention. Unfortunately these "ion pairs" often lead to limited column life. The example shown uses a Cogent Bidentate C18 column with TYPE-C Silica, which is more retentive with the simple low molecular weight " ion pair" TFA than type B phases. This phase also does not appear to suffer "phase collapse" when used in high aqueous content eluents.
This combination of properties has allowed Microsolv to suggest a simple two-step gradient,
which fulfils the "Water Soluble Vitamin" routine analytical requirements.
