Introduction

Many HPLC and LC-MS methods developed on Cogent Diamond Hydride™ columns use either acidic mobile phase modifiers, such as formic acid, or volatile ammonium salts, such as ammonium acetate or ammonium formate. While both approaches can produce excellent chromatographic results, changing repeatedly between these mobile phase systems may alter the chromatographic behavior of the column.  To ensure consistent retention, selectivity, and method reproducibility, careful consideration should be given before transitioning between acidic and ammonium salt-based methods.


Recommended Best Practice

When both mobile phase types are used routinely, it is highly recommended to dedicate separate columns for each application:

  • One column used exclusively for acid-containing mobile phases
  • One column used exclusively for ammonium acetate or ammonium formate mobile phases

This approach helps maintain consistent chromatographic performance and minimizes unexpected changes in selectivity.


Why This Is Important

Introduction of ammonium acetate or ammonium formate may modify the chromatographic characteristics of a Diamond Hydride™ column in ways that can persist over time.  As a result:

  • Retention behavior may change.
  • Selectivity may shift.
  • Method transferability may become more challenging.
  • Previously validated separations may require re-optimization.

For this reason, a column that has been conditioned with ammonium salts is generally not recommended for reuse in methods that rely exclusively on acidic additives, and vice versa.


Example of Selectivity Changes

Certain analytes may be particularly sensitive to these effects.  For example:

  • Phenylalanine
  • Phenylglycine

may exhibit reduced selectivity after a column has been exposed to ammonium acetate-containing mobile phases.

Even though retention may still occur, the separation characteristics can differ from those obtained using a column dedicated to acid-modified mobile phases.


Applications Commonly Using Acidic Modifiers

Acid-based mobile phases often contain:

  • Formic acid
  • Acetic acid
  • Trifluoroacetic acid (TFA)

These additives are frequently used for:

  • LC-MS methods
  • Pharmaceutical analyses
  • Peptide separations
  • General HPLC applications

Applications Commonly Using Ammonium Salts

Volatile ammonium buffers such as:

  • Ammonium acetate
  • Ammonium formate

are commonly employed for:

  • HILIC methods
  • ANP chromatography
  • LC-MS applications
  • Polar compound analysis
  • Metabolomics

Because these additives can influence column behavior differently than acidic modifiers, dedicated column usage is often preferred.
