Routine Use of PQ and HSQ Kit Columns

Under normal operating conditions, no special cleaning procedure is required.

Before beginning a qualification sequence:

  • Equilibrate the column for approximately 10 to 15 minutes using the recommended mobile phase.
  • Verify stable pressure and baseline conditions before starting qualification testing.

Because these columns are intended only for Performance Qualification (PQ) and Hardware System Qualification (HSQ) procedures, contamination is uncommon when they are used exclusively for their intended purpose.

When Cleaning May Be Necessary

If contamination or unexpected performance issues are suspected, the column may be cleaned using a mild flushing procedure.

Recommended Cleaning Procedure

Flush the column with:

  • 50:50 Acetonitrile / Water

for approximately:

  • 15 to 30 minutes

This procedure is generally sufficient to remove minor contamination or incidental buildup that may occur during qualification activities.

Important Usage Guidelines

To maintain qualification integrity:

  • Use the column exclusively for PQ or HSQ testing.
  • Do not use the column for routine sample analysis.
  • Store the column according to kit instructions when not in use.
  • Avoid exposing the column to unknown sample matrices or aggressive cleaning solvents.

Maintaining a dedicated qualification column helps ensure consistent and reproducible qualification results over time.

Why Dedicated Qualification Columns Matter

The columns supplied with Chemical Solutions™ PQ and HSQ Kits are selected specifically to support qualification testing.

Using these columns only for qualification work helps:

  • Maintain qualification consistency
  • Preserve established performance characteristics
  • Reduce variability in qualification results
  • Extend column service life
  • Improve confidence in instrument performance assessments

Key Takeaways

  • A 10 to 15 minute equilibration is normally sufficient before use.
  • Routine cleaning is generally not required.
  • If cleaning is necessary, flush with 50:50 acetonitrile and water for 15 to 30 minutes.
  • PQ and HSQ Kit columns should not be used for routine analytical testing.
  • Dedicated qualification columns help ensure reliable instrument qualification results.
