Introduction
Leucine and isoleucine are essential branched-chain amino acids (BCAAs) that share identical molecular weights and produce the same protonated ion at m/z 132.1025 in positive-ion mass spectrometry. Because these compounds are isobaric, chromatographic separation is required before LC-MS detection can accurately identify and quantify each analyte.
This application demonstrates the separation of D,L-leucine and D,L-isoleucine using a Cogent™ Diamond Hydride column operated under HILIC conditions. The addition of a weak acidic modifier in the mobile phase provides sufficient selectivity to resolve these structurally similar amino acids, allowing reliable quantitation. A relatively small sample volume is required, making this method suitable for serum and metabolomics applications where sample availability may be limited.
