Strong Anion Exchange (SAX) Columns for USP® L9 Methods
Silica‑Based | Always Ionized | Designed to Retain Acids
Cogent IEX SAX™ HPLC columns are strong anion exchange (SAX) columns designed for the separation and purification of acidic and negatively charged compounds in USP® L9 ion exchange methods and routine analytical workflows. These silica‑based columns are always ionized across their functional pH range, providing consistent retention and predictable performance.
Ion exchange chromatography remains one of the most widely used techniques for analyzing charged, polar molecules, and Cogent IEX SAX™ columns offer a reliable solution for laboratories requiring robust, reproducible anion exchange separations.
How Ion Exchange Chromatography Works
Ion exchange (IEX) chromatography separates charged molecules based on their reversible interaction with ionic functional groups bonded to the silica surface. Typical analytes include:
Organic acids
Amino acids
Proteins and peptides
Other polar or ionic compounds
Retention occurs through the exchange of ions between the analyte in the mobile phase and the oppositely charged ligands on the stationary phase. Retention and selectivity can be controlled by adjusting mobile phase pH and ionic strength.
The functional pH range for Cogent IEX SAX™ columns is 2.5 to 8.
Strong Anion Exchange (SAX) Selectivity
Anion exchange columns contain positively charged functional groups that retain negatively charged (acidic) analytes.
Cogent IEX SAX™ columns feature quaternary amine functional groups, which remain fully ionized at all usable pH values within the column’s operating range. This makes them ideal for methods where consistent charge interaction is required.
What “Strong” Ion Exchange Means
The term “strong” ion exchange does not refer to stronger analyte binding. Instead, it describes the ionization behavior of the functional group:
Strong anion exchange (SAX): The functional group is always ionized, regardless of mobile phase pH.
Because the charge state does not vary with pH, strong exchangers provide stable retention behavior, simplifying method development and improving reproducibility.
