Using Acid and Ammonium Salts with Cogent Diamond Hydride Columns

If your method requires both acids (e.g., formic acid) and ammonium salts (such as ammonium acetate or ammonium formate), we strongly recommend dedicating separate columns for each mobile phase type:

  • One column for mobile phases containing acids
  • Another column for mobile phases containing ammonium salts

Why This Matters

Introducing ammonium acetate or formate to a column can semi-permanently alter its chromatographic behavior for certain analytes. As a general rule, a column used for formic acid methods should not be reused for ammonium acetate methods, and vice versa.

Example:
For compounds like phenylalanine and phenylglycine, selectivity between the two decreases after ammonium acetate has been introduced to the column.
