Introduction

Analysis of dietary supplements is an important application of HPLC and LC-MS methods because product composition may not always match label claims. In some cases, compounds that are not declared on product labels may be present, making analytical verification valuable for quality control, regulatory investigations, and research applications.

This application note demonstrates the use of Cogent™ chromatography columns with LC-MS detection for the analysis of compounds found in dietary supplement products.

Overview

Certain compounds that have been associated with regulatory concern have been identified in dietary supplements. Accurate chromatographic methods are necessary to separate these compounds from complex product matrices and to provide reliable LC-MS detection.

Using appropriate chromatographic selectivity, analytes may be separated from:

  • Dietary supplement matrices
  • Plant extracts
  • Excipients
  • Other formulation components
  • Potentially undeclared ingredients

LC-MS provides both chromatographic separation and mass-selective detection, making it well suited for this type of analysis.

Chromatographic Columns Evaluated

The study utilizes:

  • Cogent™ Bidentate C18 2.o™ Columns
  • Cogent™ Diamond Hydride™ Columns

These stationary phases provide complementary selectivity and may be used for the analysis of compounds present in complex dietary supplement formulations.

LC-MS Applications

The methods described demonstrate how LC-MS analysis can be used to:

  • Detect target compounds
  • Evaluate supplement composition
  • Investigate label claim accuracy
  • Differentiate analytes from matrix components
  • Screen for additional detectable compounds

The combination of chromatographic separation and mass-spectrometric detection helps improve confidence in analyte identification.

Analytical Considerations

Dietary supplements often contain:

  • Multiple active ingredients
  • Botanical extracts
  • Flavoring agents
  • Vitamins
  • Excipients
  • Proprietary blends

As a result, analytical methods must provide sufficient selectivity to separate compounds of interest from matrix interferences.  Proper column selection can play a significant role in achieving the required chromatographic performance.
